Title: Investigation on the correlation between mechanical strain and the differentiation of mesenchymal stem cells of children's bone marrow to osteoblasts and adipocytes in vitro
Abstract: Objective To study the correlation between mechanical strain and the differentiation of mesenchymal stem cells(MSCs)of children's bone marrow tO osteoblasts and adipocytes in vitro.Methods The MSCS were isolated and primary cultured.After 3rd-6th passage of MSCs,mechanical stretch,and(or)condition medium was perform on MSCs.Cytochemical staining,RT-PCR,immunocytochemical staining and Western Blot were applied to measure the markers of osteoblasts(OC,BMP-2,I-col)and the markers of adipocytes(lipid droplets,PPARy-2).Results After mechanical stretch treatment,the level of intracellular OC mRNA was higher than that of the control group.which showed the correlation between the intracellular OC mRNA and the stimulating period.After stimulating the MSCS for 7days,the expression of BMP-2 and I-col was significantly higher than that of the control group.In the process of induction of MSCS to adipocytes,the mechanical stretched cells had less intracellular lipid droplet and lower expression of PPART-2 mRNA.Conclusions These data suggest proper mechanical strain could improve the effect on the differentiation of MSCs to osteoblasts.while inhibit the effect on the differentiation of MSCS to adipocytes.
