Methods Cell morphological assessment and MTT chromometry were both used to o...Objective This study was purposed to explore the inhibition effect and the mechanism of gambogic acid(GA) on BIU-87 cells. Methods Cell morphological assessment and MTT chromometry were both used to observe the growth inhibition of BIU-87 cells treated by gambogic acid,flow cytometry was also used to detect the apotposis and the cell cycle. Results Under inverted-microscope,typical morphological changes of growth inhibition were observed,MTT assay indicated that BIU-87 cells were significantly inhibited in growth in a dose-dependent manner and the 48 h IC50 was 1.7 μmol/L. Flow cy tometric assay showed that the apoptotic rates also was enhanced along with increasing of drug concentration,and the apoptotic rates resulting from 0,1.5,3.0 μmol/L GA were (2.2 ±0. 8)% ,(34.6 ±5.2)% and (58.6 ±6.9)% respectively. After 48 h with different concentration GA cell (0,1.5,3.0 μmol/L), G_0-G_1 stage cell numbers were (35.0 ± 1.2)% ,(35.0 ± 1.5)% ,(46.0 ±3.1)% ,and S stage cell numbers were (33.5 ±0.7)% ,(34. 1 ±0.6)% ,(15.6 ±3. 3)%. The cycle of BIU-87 was arrested in G_0/ G_1 phase. Conclusion GA could significantly inhibit the growth of BIU-87 cells,possibly through the induction of apoptosis and the arrest of cell cycle.
