Abstract: The combination of callus induction media for in vitro pineapple leaves has been developed using 2.4 D and Kinetin Plant growth regulators (PGR). This study aims to determine the effect of 2,4-D concentration, Kinetin and the interaction between both of them for induction of Sipahutar
pineapple. In vitro pineapple leaves were grown on media with concentrations of 2,4 D (0, 1, 2 ppm) and Kinetin (0, 0.5 and 1 ppm). The research design used factorial complete random designwith two way Anava. Parameters observed 1). When the callus emerges, 2). Callus color, 3)Callus biomass, 4) Callus height, 5) Callus texture, 6) Callus surface area. The results showed 1).The time of callus emergence starts on th14 day (0.5 ppm Kinetin and 2 ppm 2.4 D), until th17day (without Kinetin and 2 ppm 2,4-D). Leaves treated with MS media without 2,4 D and Kinetin
did not produce callus. 2). Callus color (observation on 28 days after induction), ranging from brown, brownish white, yellowish white to whitish green. 3). There is a significant effect of 2,4-D PGR while Kinetin has no significant effect, the interaction of both shows a significant effect on callus biomass. The highest callus biomass is 0.77 grams (1 ppm Kinetin and 2 ppm 2.4 D
treatment), the lowest biomass is 0.71 grams (0 ppm Kinetin and 2.4 D respectively). 4).
