Title: Molekularne podstawy organizacji chromatyny bakteryjnej
Abstract: Advances in high resolution microscopy techniques and development of high throughput DNA analyses allow to reconsider the views concerning bacterial chromosome (nucleoid). Recent reports show that nucleoid exhibits a hierarchical organization, similarly to the eukaryotic chromatin. However, bacterial chromosome undergoes constant modifications and topological rearrangements due to the ongoing DNA replication, transcription and translation processes. Organization of dynamic and highly compacted nucleoid structure depends on physical factors acting on chromosome molecule inside small cell compartment, and is a consequence of action of many different DNA-binding proteins. The main goal of this review is to present the recent reports on bacterial chromatin structure and to elucidate the physical and molecular factors influencing its intracellular organization.Rozwój zaawansowanych technik obrazowania (tj. mikroskopia wysokorozdzielcza) oraz wysokoprzepustowych metod analizy DNA pozwolił na zrewidowanie dotychczasowego poglądu dotyczącego struktury i organizacji chromosomu bakteryjnego (nukleoidu). Wyniki badań prowadzonych w ostatnich latach sugerują, że nukleoid, podobnie do eukariotycznej chromatyny, wykazuje hierarchiczną organizację. Jednakże, u bakterii chromosom jest pod-dawany ciągłym modyfikacjom i rearanżacjom topologicznym ze względu na jednocześnie za-chodzące procesy replikacji, transkrypcji i translacji. Organizacja dynamicznej i równocześnie silnie upakowanej struktury nukleoidu zależy od czynników fizycznych działających na czą-steczkę chromosomu w małej przestrzeni komórki bakteryjnej oraz jest wynikiem działania wielu białek wiążących się do DNA chromosomalnego. Celem niniejszego artykułu jest przed-stawienie obecnego stanu wiedzy na temat struktury chromatyny bakteryjnej oraz wyjaśnienie fizycznych i molekularnych czynników leżących u podstaw jej organizacji w komórce.
