Title: Research on the Inducement of Total Saponin of Aralia Taibaiensis to the Apoptosis in Human Leukemia Cell K562 and Its Mechanism
Abstract: Objective To study the anti-tumor effect on K562 cells and the related mechanism of Total saponin of Aralia Taibaiensis.Methods The proliferation inhibition ratio of K562 cells was evaluated by MTT test after being treated with SAT of different concentrations;Cells morphological changes after 24 hours were observed under the reverse microscope and the fluorescence microscope;The expression of protein NF-κB、Caspase-3 in K562 cells was investigated by immunohistochemistry.Result SAT inhibited the growth of K562 cells evidently in time and dose-dependent manner.K562 cells have various morphological changes about apoptosis with the increase of concentrations after 24 hours of the treatment of SAT.Compared with control group,the expression of NF-κB was down-regulated,the expression of Caspase-3 was significantly up-regulated in SAT treated group.Conclusion SAT can inhibit the proliferation of K562 cells and induce their apoptosis which may be correlated with decreasing expression of protein NF-κB and increasing expression of protein Caspase-3.
