Title: Lead Optimization in Pharmaceutical Discovery and Development
Abstract: Sensible lead candidate selection for preclinical development and registration should reduce the high rate of drug attrition, the cost of which increases with the distance that the failed drug candidate has proceeded down the research and development pipeline. This entry provides several examples of recent data on pharmaceutical development to illustrate concepts of preclinical lead optimization models and reasons for early preclinical/clinical failure, and also discusses cellular and molecular tools in regulatory toxicology.
