Title: Simultaneous analysis of flavonoids and phenolic acids in Plantago L. species using an RP-HPLC method
Abstract: A rapid reversed-phase HPLC-DAD method for simultaneous separation and determination of flavonoids (rutin, hyperosid, isoquercitrin, quercitrin, quercetin) and phenolic acids (chlorogenic and caffeic acid) in natural samples was developed and successfully applied to the analysis of Plantago L. species growing in Croatia: P. altissima L., P. argentea Chaix, P. coronopus L., P. holosteum Scop., P. lagopus L., and P. maritima L. In a single step, leaf extracts were prepared using the mixture of water and methanol (1:1, V/V), and phenolic compounds were separated by RP-HPLC with an acidified water/acetonitrile gradient within 25 minutes. Contents of analysed phenolic compounds significantly varied among examined species and were in range from 0.001% (quercitrin and caffeic acid) to 0.115% (chlorogenic acid), with reference to the dry drug. The results were evaluated by using multivariate analysis as a contribution to elucidation of chemotaxonomic relations between different taxa of genus Plantago L.
