Title: Uji Aktivitas Penghambatan Alfa Glukosidase Ekstrak Etanol 96% Kulit Pisang Ambon (Musa Paradisiaca), Pisang Kepok (Musa Balbisiana) Dan Pisang Raja (Musa Acuminata) Secara In Vitro
Abstract: Diabetes mellitus is a group of metabolic disorders characterized by an increased blood sugar levels, one of the causes of high blood sugar is α-glucosidase activity in a process of glucose formation. This research was aimed to measure the inhibitory activity of 96% ethanol extract of ambon banana (Musa paradisiaca), kepok banana (Musa balbisiana) and raja banana (Musa acuminata) peels towards the α-glucosidase inhibition which calculated from IC50 and to determine the inhibition kinetics of the extract so banana peels can be used as a herbal medicine for people with diabetes mellitus.
Assay of α-glucosidase inhibition was done by spectrophotometry method. There were five extract concentrations used in this analysis: 5, 12.5, 25, 50 and 100 µg/mL. Absorbance of samples along with acarbose, which was the positive control, was measured at λ=405 nm. Inhibition kinetics of enzyme test used the reaction system with and without inhibitor to obtain Lineweaver-Burk plot.
The value of IC50 of ambon banana, kepok banana and raja banana peels from this research were 44.764; 68.470 and 99.687 µg/mL. Acarbose was used as the positive control with IC50 = 4.479 µg/mL. The inhibition mechanism of ambon banana peels is mixed inhibition type I.
