...Objective
To etiologically diagnose and analyze two suspected patients of brucellosis and provide a etiologic basis for confirmation of the patients and control of brucellosis in Guizhou Province.
Methods
Antibody levels of Brucella in the suspected patients were determined by agar plate agglutination test and tube agglutination test. Blood culture method was used to isolate Brucella strains from the two suspected patients. Molecular and conventional techniques were used to identify suspicious bacterial strains isolated from the suspected patients.
Results
The results showed that two Brucella suspicious bacteria strains were isolated. Brucella genus specific surface protein 31 (BCSP31)-PCR showed a Brucella abortus specific 223 bp-band in both of the two bacterial strains. Brucella species-specific AMOS-PCR excluded the two strains from Brucella melitensis, Brucella suis, Brucella ovis, Brucella abortus biotype 1, 2 and 4. Conventional tests further identified the two strains as Brucella abortus biotype 3.
Conclusion
Two strains of Brucella abortus biotype 3 are firstly isolated from patients in Guizhou Province, which provides a etiological basis for diagnosis of the patients and prevention and control of brucellosis in Guizhou Province.
