Title: [Expression of metastasis tumor-associated protein 2 in cervical squamous carcinoma and its relationship with prognosis].
Abstract: To investigate the expression of metastasis tumor-associated protein 2 (MTA2) in cervical squamous carcinoma and its relationship with prognosis. Methods: Immunohistochemistry and real-time PCR were performed to determine the expression and distribution of MTA2 mRNA and protein in normal cervical tissue, cervical intraepithelial neoplasia (CIN) and cervical squamous carcinomas tissues, then its relationship with clinical pathological factors and prognosis was analyzed. Results: The positive rate of MTA2 protein in normal cervical tissue, CIN and cervical squamous cell carcinomas tissues were 0, 30.0%, 73.4%, respectively. The positive rate was associated with international federation of gynecology and obstetrics (FIGO) stage and lymph node metastasis, whereas there was no correlation with the age of patients or the degree of tumor differentiation. The expression of MTA2 mRNA in normal cervical tissue, CIN and cervical squamous carcinomas tissues was 0.437±0.028, 0.737±0.102 and 1.172±0.068, respectively. The positive rate was associated with FIGO stage and lymph node metastasis, whereas there was no correlation with the age of patients or the degree of tumor differentiation. The result of survival analysis showed poor overall survival time in the patients with high expression of MTA2. Multivariate COX proportional hazards model showed that the positive expression of MTA2 protein, FIGO stage and the metastasis of lymph node were independent prognostic factors for unfavorable clinical outcome of cervical cancer. Conclusion: The positive expression of MTA2 was closely related to the development, invasion and metastasis of cervical squamous cell carcinomas. The positive expression of MTA2 protein, FIGO stage and the metastasis of lymph node were independent prognostic factors for unfavorable clinical outcome of cervical cancer. The expression of MTA2 could be used as a potential molecular marker in evaluating the prognosis of cervical squamous cell carcinomas.目的:研究转移相关基因2(metastasis tumor-associated protein 2,MTA2)在子宫颈鳞癌组织中的表达水平,探讨其与子宫颈鳞癌临床病理因素之间的联系及其预后价值。方法:采用免疫组织化学和RT-PCR检测MTA2蛋白及mRNA在子宫颈鳞癌、子宫颈上皮内瘤变及正常宫颈组织中的表达情况,分析其与临床病理因素的关系,同时综合临床病理资料及随访数据对子宫颈鳞癌患者进行预后分析。结果:MTA2蛋白在正常宫颈组织、子宫颈上皮内瘤变组织、宫颈鳞癌组织中的表达率分别为0,30.0%,73.4%,且其表达水平与宫颈癌国际妇产科协会(International Federation of Gynecology and Obstetrics,FIGO)分期、淋巴结转移有关(P<0.05),而与患者年龄及肿瘤的分化程度无关(P>0.05);MTA2 mRNA 在正常宫颈组织、子宫颈上皮内瘤变组织、宫颈鳞癌组织中的表达量分别为0.437±0.028,0.737±0.102,1.172±0.068,且其表达量亦与宫颈癌FIGO分期、淋巴结转移有关(P<0.05),而与患者年龄及肿瘤的分化程度无关(P>0.05);Kaplan-Meier法进行单因素生存分析,MTA2蛋白表达阳性患者与阴性患者无瘤生存期分别为(23.19±1.87)和(39.57±3.16)个月,经log-rank检验得出两组患者无瘤生存期差异有统计学意义(χ2=20.225,P<0.001);多因素COX比例风险模型预后分析显示,MTA2蛋白表达水平、FIGO分期及淋巴结转移为宫颈癌有效的独立预后指标(P<0.05)。 结论:MTA2的高表达与宫颈癌的发生、进展及转移有关;MTA2蛋白表达水平、FIGO分期及淋巴结转移可作为宫颈癌预后的3个相对独立的因素;MTA2有可能成为判断宫颈癌患者进展及预后的一个新的分子标志物。.
