Title: Susceptibility of Enterococcus faecalis biofilm in vitro
Abstract: Objectives To observe the structure of Enterococcus faecalis(E.faecalis) biofilm by an experimental biofilm model in vitro.To observe the susceptibility of 24-hour E.faecalis biofilm to calcium hydroxide and chlorhexidine and estimate the efficacy of two medications against E.faecalis biofilms.Methods E.faecalis biofilms were formed on glass slices at 4 h,8 h,12 h,24 h and 48 h,respectively.E.faecalis biofilms formed on glass slices at 24 h were cultured with 10% calcium hydroxide and 2% chlorhexidine for 3 min,10 min or 30 min.Biofilms observed by CLSM.The data were analyzed with One-Way ANOVA.Results It was shown that E.faecalis biofilm had three-dimensional spatial structure with various forms.As for the biofilms within 48 h,the percentage of vital bacteria increased from the inner layer to the outer.Chlorhexidine was more effective than calcium hydroxide against E.faecalis biofilm at 3 min,but there were no significant difference between calcium hydroxide and chlorhexidine after 10 min.Conclusions E.faecalis biofilm had a certain degree of thickness.Its structure was three dimensioned,heterogeneous,openning and of various forms.When E.faecalis biofilms were exposed to 10% calcium hydroxide and 2% chlorhexidine for 30 min,most of the bacteria were killed.
