Title: Application of the MTT colorimetric assay to comparison of the cellular growth curve of the colorectal and cystic cancer.
Abstract: Objective To study the best experimental condition for chemosensitivity of human colorectal and cystic cancer cell line in vitro. Methods To measure the cellular growth curve using MTT colorimetric assay, in order to realize the logarithmic growth time of the two kinds of cell (the best experimental intervention time) and the prime density of cultivation. Results The best measure condition for the cell strain-174 of fast multiplication colorectal cancer was at a wavelengh of 630 nm, but the wavelengh of 450 nm is better measure condition for the cell strain of slow cultivation cystic cancer. Conclusion The former's best inoculation density is 1.5~5.0×10~4cells/ml, logarithmic growth time is the 3th~5th day after inoculation; but the catter's perfect density is 1.4~2.6×10~4cells/ml, logarithmic growth time is the 3th~7th day after inoculation.
