Title: Extracted methods of arsenic speciation in liver of rats treated with Sodium arsenite
Abstract: Objective To investigate the extracted methods of arsenic speciation in liver of rats treated with Sodium arsenite,to establish the determination means of arsenic speciation and to understand further the basic data about relationship between it's metabolism and mechanism of toxicity in vivo.Methods The arsenic species in organs of rats in sodium arsenite high,middle and low groups were respectively extracted using accelerated solvent extraction(ASE),sonication extraction(SON) and hydrochloric acid(HCl) extraction,and applied the high efficiency liquid chromatography and hybrid genesis atomic fluorescene spectroscopy(HPLC-HGAFS) to determine arsenic speciation and total arsenic.Results The extraction rate of ASE was higher than SON and HCl.The extraction rate of SON was higher than HCl;and the extracted and quantitative methods of arsenic speciation of sodium arsenite(iAS3+) and dimethylarsinic acid(DMA) were established.The average recovery rates of DMA was 98.9%~102.9%.Conclusion The extraction rate of extrated arsenic speciation of ASE is better than the SON and HCl,DMA is the main species in organs of rats treated with Sodium arsenite.
