Title: Influence of pretreatment with dexmedetomidine on hemeoxygenase-1 in rats with lung injury induced by limb ischemia-reperfusion
Abstract: Objective To investigate the effects of pretreatment with dexmedetomidine on hemeoxygenase-1 in rats with lung injury induced by limb ischemia-reperfusion. Methods Thirty healthy SD rats weighing 250-300g were randomly assigned into 3groups(n=10,each one):sham operation(sham group),limb IR group(IR group) and dexmedetomidine group(Dex group). Dex group received iv dexmedetomidine 25μg / kg slowly at 10 min before ischemia.In sham and IR group the animals received equal volumes of NS instead of dexmedetomidine. The rats were sacrificed at the end of reperfusion and the lungs were immediately removed for pathological examination and determination of the lung Wet / dry weight ratio(W / D). The arterial blood was taken for blood gas analysis. The level of hemeoxygenase-1 as detected by ELISA. Results Compared with sham group,in IR group and Dex group PaO2and PaCO2was decreased;W / D and HO-1 content was increased,and lung damage was also appeared(P 0.05); compared with IR group,the damage of lung was significantly reduced in Dex group;PaO2and PaCO2was increased;W / D was decreased and HO-1 level was increased(P0.05). Conclusion Dexmedetomidine pretreatment can improve the activity of HO-1,up-regulate the expression of HO-1 protein,so that relieve lung injury induced by ischemia-reperfusion.
