Title: Artesunate induce the apoptosis of HepG2 and mechanism
Abstract: Objective To investigate the roles of artesunate on the apoptosis of human hepatoma cell lines HepG2,also to explore its possible mechanism of anticancer effects.Methods Human hepatoma cell lines HepG2 cultivated by convention,were randomly divided into 2 groups:untreated control group;different concentration artesunate treated with HepG2.After treated with different concentration for 48 h,apoptosis was assessed by flow cytometry together with Wright ' s staim.RT-PCR method was used to detect caspase-3 expression at the mRNA level.Results Exposed to artesunate(3.125 μg/ml,6.25 μg/ml,12.5 μg/ml,25 μg/ml,50 μg/ml) for 48 h,the apoptosis of HepG2 cells was accelerated in a concentration-dependent manner.Light microscopy revealed early apoptotic changes of the cells with obvious chromatin condensation by Wright ' s staim.RT-PCR showed that the expression of caspase-3 mRNA in HepG2 cells was significantly increased.Conclusions Artesunate can induce the apoptosis of HepG2 by mechanism related to caspase-3 pathway.
