Title: Influence Factors of Transplanting Survival Rate of Tissue-cultured Seedlings of Water Iris
Abstract: [Objective] The purpose of the study was to provide technical basis for the factorial production of water iris seedlings.[Method] With the tissue-cultured seedlings of water iris as tested material,the influence factors of transplanting survival rate of tissue-cultured seedlings were studied through transplanting experiment.[Result] The survival rate of tissue-cultured seedlings transplanted into the substrate peat + rice chaff ash + perlite(Their volume ratio was 7∶2∶1.)was above 90% and that of tissue-cultured seedlings transplanted into the other 2 substrates were below 60%.The average root number,leaf number and main root length of transplanted tissue-cultured seedlings from rooting medium 1/2 MS+IBA0.5 mg/L+NAA 0.5 mg/L+active carbon(Treatment A3)were significantly superior to that of transplanted tissue-cultured seedlings from 1/2 MS media and the transplanted seedlings from 1/2 MS medium and without black pollution on their root crowns(Treatment A2)were significantly higher than that from the other 2 treatments.The transplanting survival rates of big and middle tissue-cultured seedlings from Treatment A2 and A3 were above 95%.The survival rates of tissue-cultured seedlings transplanted on Apr.13th and Sep.9th were highest and they were 81% and 82% resp.The seedling formation time of tissue-cultured seedlings transplanted on Jun.2nd was shortest and it was 36 d.The survival rate of tissue-cultured seedlings transplanted after Nov.3rd was low and their seedling formation time was prolonged to 120 d.[Conclusion] This experiment laid a foundation for the cultivation and breeding of water iris.
