Title: Preventive effects of Zafirlukast on colorectal cancer induced by azoxymethane in rats
Abstract: Objective Zafirlukast is a peroxisome proliferator-activated receptor gamma(PPARγ) ligand.The aim in this study is to explore the chemopreventive effects of Zafirlukast on the formation of aberrant crypt foci(ACF) of rat's large intestine and colorectal cancer induced by azoxymethane(AOM).Methods Colorectal cancer of rat was induced by AOM intraperitoneal injection.After adaptive feed for 1 week,80 Wistar rats were randomly divided into three groups:control group(ordinary diet feed,n =20),AOM group(ordinary diet + AOM inducing cancer,n = 30) and prevention group(Zafirlukast feed plus AOM inducing cancer,n = 30).The numbers of ACF and aberrant crypt(AC) in large intestine were observed by methylene blue staining at 12th week of experiment,and the incidence of colorectal cancer were calculated and compared at the end of experiment.Results At 12th week of experiment,the ACF were found in AOM group and prevention group,and the average number of ACF and AC in prevention group were lower than those in AOM group(all P 0.05) ;In control group,the ACF were not found.At 32th week of experiment,the colorectal cancers were detected in AOM group and prevention group.The incidence of colorectal cancer was 72.22%(13 /18) in AOM group against 42.11%(8 /19) in prevention group,and there was significant difference in incidence of colorectal cancer between prevention group and AOM group(P 0.05).Conclusions Zafirlukast can decrease the number of ACF and AC in the early stage of experiment and significantly prevent the production of colorectal cancer induced by AOM.
