Title: Study on an amylase-producing actinomycetes from ocean:isolation,identification,mutation and optimization of culture conditions
Abstract: To obtain the actinomycetes which could produce amylase,the selective media were used.The strains with relatively high-producing amylase were screened through measuring enzyme activity by mean of Yoo.In order to improve the ability of producing amylase many methods were used,such as UV mutagenesis,ultrasonic mutagenesis and heat mutagenesis.In addition,culture conditions of the strain were optimized by using single-factor and orthogonal tests.At the same time,its identification was done through the morphology,colony characteristics,physiological and biochemical characteristics and the analysis of 16S rDNA homology.43 strains producing amylase were isolated from marine mud and marine algae collected from the coastal regions of zhoushan(in Zhejiang province),among them 3 strains had the ability of high-producing amylase.Treatment with UV mutagenesis the enzyme activity of strain A24 increased to 282.7 U·mL-1 from 111.5 U·mL-1,and then by optimization of culture conditions to 353.67 U·mL-1.The strain was identified as Streptomyce sp.A24 based on its morphological and physiological properties,and 16S rDNA sequence analysis.GenBank accession number was GU184337.
