Title: Cloning of IFIT1 Gene and Its Bioinformatics and Tissue Expression Analysis in Swine
Abstract: The cDNA of IFIT1 gene was cloned with RACE(rapid amplification of cDNA end)and its structures and functions were analyzed by bioinformatics.The mRNA distribution of the porcine IFIT1 was analyzed in 11 tissues(lung,heart,kidney,tongue,bladder,ovary,spleen,liver,skeletal muscle,stomach and small intestine) of newly-born and 1-month-old piglets with RT-PCR.The result showed that the sequence of IFIT1 was 1 971 bp(GenBank ID: HQ679904) in length,containing 48 bp 5' UTR and coding region(CDS,1 437 bp) and 487 bp 3' UTR and encoding 478 amino acids.The isoelectric point(PI) of the protein was 6.18,and the molecular weight was 55 232.9×103.Its secondary structures were shown to be mainly α-helix and Coli and the tertiary structures contained multiple repeats of tetratricopeptide repeat helix-turn-helix motifs.Real-time PCR results indicated that the expression level of porcine IFIT1 differed significantly among tissues of the newly-born piglets and the one-month-old piglets(p0.01) and IFIT1 gene showed the highest expression levels in spleen and skeletal muscle in the two periods,respectively(p0.01).The above results may lay a foundation for identification and application of IFIT1 gene in further research.
