Title: Study of the association between VKORC1-1173C>T polymorphism and warfarin maintenance dose
Abstract: Objective:To investigate the association between VKORC1 1173CT polymorphism and inter-indi-vidual warfarin maintenance dose.Method:The genotypes of 323 patients with stable warfarin dose were determined by using of real-time PCR combined with melting curve analysis.The warfarin maintenance doses were compared among patients with different genotypes.Result:The differences in genotype distributions and allele frequencies of VKORC1 1173CT between Han people and minority ethnic group in Guizhou province have no statistical significance.In the total of 323 patients,the numbers of TT,TC and CC genotype were 253,66 and 4,with the frequencies of 78.3%,20.4% and 1.2%,respectively.The frequencies of T and C allele were 88.5% and 11.5%,respectively.Among the 3 genotype groups,patients with TT genotype exhibited a mean stable warfarin dose of 2.58±0.78 mg/day.This dose was significantly lower(P0.001) than TC heterozygotes and CC homozygotes,in which the mean stable doses were 4.25±1.35 and 4.53±0.94 mg/day,respectively.Conclusion:VKORC1 1173CT polymorphism is a major genetic factor that influence the inter-individual warfarin maintenance dose,and determination of VKORC1 1173CT genotype could be used for personalized medicine in warfarin therapy.
