Title: SSR fingerprints and genetic variations of the 32 persimmon major cultivars.
Abstract: The aim of this study was to construct a DNA fingerprinting database of 32 persimmon major cultivars,and the genetic diversity was analyzed based on simple sequence repeats(SSR).Twenty core SSR primer pairs with high polymorphisms and good repeatability were successfully screened from 78 candidates.183 polymorphic bands were amplified from the 32 cultivars by 20 primer pairs,and 9.15 genotypes were detected by each SSR primer pair on an average ranging from 3 to 20.The level of polymorphism was 81.3%,the heterozygosity of each locus ranged from 0.410 3~0.914 3,with an average level of 0.702 7.Twelve cultivars had specific genotypes by 8 primer pairs.32 cultivars could be identified by 5 primer combinations at least.Due to the complex SSR patterns in persimmon,it was difficult to score each band with number 1 and 0.Here,we scored each SSR pattern with an alphanumeric code instead of with pairs of 1 and 0.By arranging all of pattern numbers,we could build the fingerprint code for every persimmon accession.Core primer combination method is better than distinct band method for the construction of DNA fingerprinting of persimmon major cultivars in China.The cultivars used in this study were divided into 2 groups by the cluster software of NTSYS-pc V2.10,which indicated that the genetic relationship of the persimmon cultivars derived from the same district is similar to a certain extent.
