Title: Effect of Inactivate Synovial Fluid Cell from Rheumatoid Arthritis Patients on IFN-γ Production of Activate Synovial Fluid Cell
Abstract: Objective To observe the effect of inactivate synovial fluid cell from rheumatoid arthritis patients on IFN-γproduction of activate synovial fluid cell.Methods Synovial fluid in the knee joint of RA patients selected was drawn,then isolated the synovial fluid cells,which were randomly divided into treatment group and control group.In the treatment group,2.0×10~6/mL activate synovial fluid cells were mixed cultured separately with 2.0×10~5/mL、4.0×10~4/mL、2.0×10~4/mL inactivate synovial fluid cells,the ratio of inactivate synovial fluid cell to activate synovial fluid cell to be correspondingly 1:10、1:50、1:100.In the control group,2.0×10~6/mL activate synovial fluid cells were cultured purely.All above cells under the condition of 37℃and 5%CO_2 were cultivated for 48 hours.At last,the content of IFN-γwas determined by ELISA.Results(1) The IFN-γcontent was decreased obviously in the treatment groups compared with control groups(P0.05).(2) The IFN-γlevel was significantly lower in the 1:10 group than that in the 1:50 and 1:100 group.There was no obvious difference about IFN-γbetween 1:50 and 1:100 group.Conclusion There might be a depressant effect of inactivate synovial fluid cell on IFN-γexcreted by synovial fluid cell in the rheumatoid arthritis patients.The depressant effect should be the strongest,when the ratio of inactive synovial fluid cell to active synovial fluid cell is 1:10.The mechanism shoul need to be investigated still further.
