Title: The changes of nuclear factor-κB(NF-κB) in myocardial ischemic/reperfusion injury
Abstract: Objective To study the changes of nuclear factor-kappa B(NF-κB) and cytokines in myocardial Ischemic/Reperfusion injury during cardiopulmonary bypass(CPB).Methods Twelve healthy male dogs(21~28 kg) were randomized divided into Methylprednisolone group(group M,n=6) and control group(group C,n=6).CPB was performed with cardiac arrest by cardioplegic solution during aorto-block.Cardiac resuscitation was made after 120 min of aorto-block and weaned from CPB when circulation lasting.Blood-serum cytokines(TNF-α,IL-1β,IL-6,IL-8),NF-κB P_(65) and pathology change of myocardium tissue were measured before CPB(T_0),before release of aorto(T_1),30 min(T_2) and 90 min(T_3) after release of aorto.Results The activation of NF-κB,levels of all cytokins increased significantly at T_1,T_2,T_3 than that of at T_0 in two groups(P0.01).The activation of NF-κB,levels of all cytokins decreased significantly in M group than that of C group at T_1,T_2,T_3(P0.01).Tissue section display that neutrophil accumulated in myocardium tissue increased at T_1,T_2,T_3 in two groups and M group was slighter than C group.Conclusion Myocardial Ischemic/Reperfusion injury was involved with the activation of NF-κB which increased the level of the cytokines.Methylprednisolone strongly attenuates the activation of NF-κB and the level of the inflammatory cytokines,protect the heart from Ischemic/Reperfusion injury during CPB.
