Title: Analysis of gene chip on acute arsenite-induced and chronic arsenite-induced L-02 Cells
Abstract: Objective c DNA microarrays were used to investigate the expression profile of human genes in response to acute and chronic sodium arsenite treatment.Methods The RNA was purified from the L-02 cells without and with arsenite sodium induction for 2,15,24h and 2 weeks respectively.Results The hybridization patterns were quite different between acute and chronic sodium arsenite treatment.The genes related with detoxification function and protein synthesis function were activated at the acute arsenite induction.However after long-term exposure to arsenite,tumor-related genes were expressed highly,while immunity-related genes were inhibited.Conclusion The genes related with detoxification function were activated at the acute arsenite induction.However,under long-term arsenite exposure,the differentially expressed genes could give a clue to carcinogenesis of arsenite.The ways of resistance to arsenite toxicity were different between acute and chronic sodium arsenite treatment.
