Title: Effet of Kangxian Fufang-I on the initiation of hepatic stellate cells in rats with alcoholic liver disease
Abstract: Objective:To investigate the effect of Kangxian Fufang Ⅰ(KXⅠ)on hepatic stellate cells in rats with ethanol-induced liver disease.Methods:Wistar rats weighing between 160g and 200g were intragastricly fed with ethanol(8 to 10g·kg-1·d-1)or ethanol plus KXⅠ(4.0ml·kg-1·d-1)for 12 weeks.Control rats received normal sodium instead of ethanol.At the end of 4,8,12w,rats were killed and received liver histopathology and ultrastructure test.Hepatic hyaluronic acid(HA)and laminin(LN)were measured by radioactive immunoassay.Results:Up to 12 weeks,marked hepatic fatty changes,inflammation,necrosis and fibrosis were observed in ethanol-fed rats.In contrast,these alternations were attenuated by KXⅠ administration although mild fatty changes remained.By electron microscope,liver ultrastructure was observed that hepatic stellate cells(HSC)increased,became myofibroblast-like cells and there was a large of collagen around cells in ethanol-fed rats whereas HSC remains normal shape and only a little of collagen was observed in medicine-treated rats.At the end of 12 weeks,hepatic HA and LN reached the highest levels in ethanol-fed rats and were significantly higher than controls and medicine-treated groups(74.85±9.23 vs 41.26±8.34、53.62±4.85,P0.01;37.69±5.01 vs 22.34±3.51、28.07±4.11,P0.05).Conclusion:KXⅠ decreased the levels of hepatic HA and LN,attenuated ethanol-induced hepatic fibrosis in rats.The mechanism was contributed to inhibit hepatic stellate cells initiation.
