Title: The mechanism of hyperin on cell cycle arrest and apoptosis of human breast cancer cell
Abstract: Objective: To investigate the effect and mechanism of the Hyperin on cell cycle and apoptosis on human breast cancer cell lines MCF-7. Methods: MCF-7 cells cultured in vitro,MTT assay was used to detect cell viability with various concentration of Hyperin,flow cytometry was used to detect cell cycle,western blot to test protein expression of cell cycle protein and apoptosis retelated protein. Results: Compared with vehicle treated,cell viability decreased in a dose and time dependent way treated with Hyperin,and cell cycle was blocked in G2 / M phase and cell number in G2 / M phase was increased and S phase was obviously decreased at the meantime,expression of cyclin-dependent kinase 2( CDK2),Cyclin A was decreased and caspase 3 were significantly increased compared to control group. Conclusion: By activating the cell cycle arrest related apoptosis pathway may be one of the mechanism of antitumor effect of Hyperin.
