Title: Effects of Resveratrol on Apoptosis of Myocardial Tissue in Rabbits with Atherosclerosis
Abstract: Objective To investigate the effect of resveratrol on myocardial mitochondria-mediated apoptosis in rabbits with artherosclerosis.Methods Seventy male New Zealand white rabbits were randomly divided into 5 groups:A for normal control group,B for pathological control group,C for pathological control group treated with low dosage resveratrol(4 mg·kg-1·d-1),D for pathological group treated with mid dosage resveratrol(8 mg·kg-1·d-1) and E for pathological group treated with high dosage resveratrol(16 mg·kg-1·d-1).At the end of the 12th week,left ventricular tissues were analyzed by histopathological examination,and apoptotic cell were detected by Terminal deoxynucleotidyltransferase-mediated dUTP nick end labeling.The mRNA expression of bax and bcl-2 were detected by semiquantitative reverse transcriptase polymerase chain reaction.Results There were no obvious morphological changes in each group during H-E staining.Apoptotic index in group B was significantly increased when compared with group A,C,D and E(P0.01).Resveratrol can inhibit apoptosis of myocytes in a dose-dependent manner(P0.05).The mRNA expression of bax and bcl-2 were significantly increased in group B than in group A(P0.01).The mRNA expression of bax was significantly decreased after resveratrol intervention,simultaneously the mRNA expression of bcl-2 was increased(P0.01).Conclusions Resveratrol can promote expression of the anti-apoptotic gene bc1-2 and decrease the pro-apoptotic gene bax in myocardial cells of rabbits with atherosclerosis.Resveratrol may inhibits myocardium apoptosis by reducing bax/bcl-2 ratio.
