Title: cDNA cloning and sequence analysis of S1 segment of new isolated reovirus strains
Abstract: Objective: To clone,sequence and analyze genome segment 1(S1) of new isolated reovirus strains.(Methods:) Virus genome was extracted from L929 cell infected with the isolate.Full-length cDNA clone of S1 segment was obtained by improved single primer amplification technique.Nucleotide and amino acid sequences were analyzed using free computer softwares.Results: The total nucleotide length of S1 segment was 1 437 bp.Nucleotide homology with 3 prototypic strains T1L,T2J and T3D was 59%,61%,and 48%,respectively.Amino acid homology of deduced sigma1 protein with 3 prototypic strains was 52%,60%,and 26%,respectively.Conclusion: S1 segment of new isolated reovirus is remarkably different from that of other known strains,and it might belong to an independent branch of serotype 2 reovirus in evolutionary tree.
