Title: Determination of Quercetin and Rutin in Lespedeza virgata with RP-HPLC
Abstract: Objective To set up an RP-HPLC method for the separation and determination of quercetin and rutin in Lespedeza virgata. Methods Stationary phase: Lichrosorb C_(18) column(4.6 mm×150.0 mm,5 μm);mobile phase : acetonitrile methanol-water-phosphoric acid(47∶53∶0.2);flow rate :1 mL·min~(-1);detection wavelength: 371 nm;column temperature:40℃. Results The calibration curves were linear in the ranges of 0.25-2.01 μg·mL~(-1) for quercetin and(2.54-20.32) μg·mL~(-1) for rutin.The average rates of recovery of quercetin and rutin were 98.7%(RSD=1.26%) and 99.2%(RSD=1.07%),respectively. Conclusion The method was shown to be rapid,accurate and well reproducible.
