Title: Dynamic expression of nuclear factor-kappa B and ICAM-1 in the pathological process of bleomycin-induced pulmonary fibrosis in mice
Abstract: Objective To investigate the action of nuclear factor-κB(NF-κB) and intercellular adhesion molecule-1(ICAM-1) in the lung tissues of mice in the progress of bleomycin-induced interstitial pulmonary fibrosis(IPF).Methods Pulmonary fibrosis model was induced by intratracheal instillation of single dose bleomycin(BLM) 5mg/kg in male Wistar rats.Dexamethasone was administered once every other day in the therapeutic groups.The animals were killed on the 1th,3th,7th,14th and 28th day separately.Hold the lung tissue,stain it with HE,investigate the lung tissue pathological changes.Detect the contents of p65 and ICAM-1 in the lung tissue by using the method of immunohistochemistry.Results Compared with NS group,NF-κB and ICAM-1 in lung tissue in BLM group and therapeutic group were obviously higher.The expression of NF-κB and ICAM-1in lung tissue in BLM group were separately the highest at the 1th day,the 7th day,then dropped in.Compared with those of BLM group,the expression of NF-κB and ICAM-1 in the lung tissue in therapeutic group were lower.Conclusion NF-κB and ICAM-1 may play an important role in the early stage of IPF,and the enhanced expression of NF-κB and ICAM-1 may be involved in pulmonary fibrosis.
