Title: Effect of HER-2 siRNA on the drug resistance of ovarian carcinoma SKOV-3 cells to cisplatin
Abstract: Objective: To study the effect of HER-2 siRNA on the drug resistance of ovarian carcinoma SKOV-3 cells to cisplatin.Methods: HER-2 siRNA was synthesized in vitro and then transfected into SKOV-3 cells.The expression of HER-2 mRNA both before and after transfection was detected by RT-PCR.The chemosensitivity of transfected and untransfected cells to cisplatin was detected by MTT method.Results: Sequence specific siRNA targeting HER-2 down-regulated the expression of HER-2 mRNA markedly 72 hours after the transfection.In the presence of different concentrations of cisplatin,there were significant differences both between HER-2 siRNA transfected group and non-specific siRNA transfected group and between HER-2 siRNA transfected group and control group(P0.01).The chemosensitivity of HER-2 siRNA transfected cells to cisplatin increased by 10 times.Conclusion: Sequence specific siRNA targeting HER-2 could inhibit the expression of HER-2 in SKOV-3 cells and thus increase the sensitivity of the cells to cisplatin.RNA interference provides a new strategy to the treatment of ovarian carcinoma.
