Title: Effects of different magnitude mechanical strain on alkaline phosphatase activity and osteocalcin of mouse osteoblast-like cells(MC3T3-E1) in vitro
Abstract: Objective:To investigate the effects of different magnitude mechanical strain on alkaline phosphatase(ALP) activity and osteocalcin of mouse osteoblast-like cells(MC3T3-E1).Method:MC3T3-E1 cells were subjected to 6%,12%,18% elongation by using multi-passage cell stress loading system simultaneously at different intervals(24 h,48 h).Biochemical analysis and radio-immunological methods were used to study the effects of different magnitude mechanical strain on alkaline phosphatase(ALP)activity and expression of osteocalcin(OCN) respectively.Result:The ALP activity of MC3T3-E1 cells significantly decreased 24h and 48h after mechanical strain treatment with stretching force increasing(P0.01);however,the expression of OCN significantly increased only at 12% elongation(P0.01),there was no significant difference among other groups(P0.05).Conclusion:Different magnitude mechanical strain can affect ALP activity and osteocalcin of MC3T3-E1 cells in strain-dependent manner.
