Title: Establishment of bladder cancer cell line expressing enhanced green fluorescent protein gene
Abstract: Objective To establish the bladder cancer line stable expressing enhanced GFP gene and observe its biological characteristics.Methods KU-7 cell line was transfected by enhance GFP(EGFP) gene in liposome-mediated gene transfer and selected by G418.The expression of EGFP in infected cells was observed by fluorescence microscopy and detected by flow cytometry.Multiple biological behaviors,such as the growth curve and cell adherent rate were compared between the infected and uninfected cells.EGFP expression in subcutaneous tumors was assessed in vivo.Results After being transduced,bladder cancer KU-7 cells with the EGFP gene displayed a stable and long-term EGFP expression under the nonselective conditions in vitro.Their biological behaviors,such as the growth curve and cell adherent rate were not different compared to untransfected parental cells in vitro.In subcutaneous tumors,EGFP was stable and highly expressed.Conclusion The bladder cancer cell line stable expressing EGFP has been successfully established.
