Title: EFFECTS OF CALCIUM ANTAGONIST ON APOPTOSIS AND EXPRESSION OF bcl-2,bax IN RENAL TISSUES OF RAT WITH TYPE 2 EXPERIMENTAL DIABETES
Abstract: ObjectiveTo investigate apoptosis and the expression of apoptotic regulatory proteins bax and bcl-2 as well as the effects of calciumr antagonist on apoptosis and the expression of apoptotic regulatory proteins in the kidneys of rats with type 2 experimental diabetes.MethodsMaking 2 type diabetic rats mode by given high sugar and high fatdiets,associating injection of small count streptozotocin(STZ) in abdominal cavity.The animals were divided into three groups: group control, group diabetic and group treatment group six animals were killed respectively at weeks 8,12 after STZ injection.The kidncy wasremoved and prepared for light microscopy,immunohistochemistry,flow cytometry.ResultsImmunohistochemically,the staining reactions of kidney for bax and bcl-2 were stronger at group diabetic and group treament than group control.flow cytometry analysis showed the rate of apoptosis and the expression of bcl-2 and bax increase in type 2 diabeic nephropathy rats.CCB can reduce the rate of apoptosis,and the expression of bax,but it can promote the expression of bcl-2 in type 2 diabeic nephropathy rats.ConclusionMaking type 2 diabetic rat mode by given the high-sugar and high-lipid diets associating injection of small count STZ,FBG、Ins、TG、TC、Uaer all significantly increased in diabetic group,and as the progression of diabeic nephropathy,they were more severe.Apoptosis may be involvedin pathological change in type 2 diabeic nephropathy rats.The effect of amlodipine to improve renal function may be partly through influencing renal cells apoptosis and the expression of apoptotic regulatory proteins in type 2 diabeic nephropathy rats.
