Title: APOPTOSIS OF HEPTOMA CELL LINE HepG2 INDUCED BY IP_6 AND ITS EFFECTS ON EXPRESSIONS OF PROTEINS Bcl-2 AND Bax
Abstract: Objective To explore the effects of IP6 on apoptosis of heptoma cell line HepG2 induced by HepG2 and expressions of Bcl-2 and Bax proteins. Methods HepG2 cells cultured in vitro were exposed to various concentrations of IP6.Thereafter,the cellular morphological changes were observed under fluorescence microscope with Hoechst 33258 staining.The effect of IP6 on apoptosis of HepG2 cells was tested by flow cytometry.The expressions of Bcl-2 and Bax were examined by immunocytochemical staining. Results The apoptotic changes in HepG2 cells exposed to IP6 were observed under fluorescence microscope.The apoptosis rate of HepG2 cells treated with IP6 increased and it correlated positively with concentration of IP6(F=342.15,q=2.98-28.53,P0.05).IP6 decreased the expression of Bcl-2 protein(F=110.21,q=5.88-16.92,P0.05) and strongly increased the expression of Bax(F=26.00,q=2.97-8.19,P0.05) at the concentration of 1.0,2.0 and 3.0 mmol/L.Conclusion IP6 induces the apoptosis of HepG2 cells is probably by regulating the expressions of Bcl-2 and Bax.
