Title: Isolation and Physiological Characterization of a Marine Psychrotroph Pseudoalteromonas flavipulchra HH407 Producing Cold-active Alkaline Protease
Abstract: A strain HH407 which could production cold-active alkaline protease,was isolated from the sea area of Lianyungang.Then,the identification and physiological characterization of the strain HH407 were studied.The results showed that the strain HH407 was gram negative,rod shaped,aerobic,with a single polar flagellum,neither endospore nor capsule formed,about 0.4~0.7μm×1.0~1.8μm in size.The colony on screening medium were circular,yellow color, smooth,translucent,convex with regular edges.Based on the 16S rDNA sequence and phylogenetic analysis,as well as the physiological and biochemical tests,the isolate as classified to Pseudoalteromonas flavipulchra HH407.The strain was facultative psychrotrophs by the growth condition test with the optimum growth about 20℃and grow at 0~40℃.The optimal pH for growth was 8.5 with pH range for growth between pH 5.5 and pH 11.It can grow at a wide range of NaC1 concentration of 0.5~13 g/dL with the optimal at 4 g/dL,no growth without NaC1.The protease produced by HH407 exhibited the high activity at pH10.0 and 35℃,respectively.
