Title: Microfertilization by epididymal sperm of mouse
Abstract: Aim: To establish the methods of microfertilization and explore the fertilizing potential of mouse sperm obtained from caput epididymidis and cauda epididymidis. Methods: ICSI and SUZI were employed to obtain the assisted fertilization of mouse sperm from caput epididymidis and cauda epididymidis. Results:Single epididymal sperm was injected intracytoplasmically(ICSI) into mouse oocytes. Of 14 survival ooctyes injected with cauda epididymal sperm, 5 oocytes were fertilized and developed to 2-cell stage. Whereas 25 oocytes survied the intracytoplasmic injection of single caput epididymal sperm, 9 of which fertilized and cleaved to 2-cell stage. Subzonal insemination(SUZI) was carried out by injection of 1-3 cauda epididymal sperm into perivitelline space of mouse oocyte. 4 out of 30 SUZI oocytes fertilized. Conclusion: These results demonstrate that we have succeed in the fertilization of mouse oocytes by micro-injection of epididymal sperm, but the fertilization rates remain to be improved. [
