Title: Triptolide induces apoptosis of human pancreatic cancer cell line panc-1 cells in vitro
Abstract: Objective To study the effect of triptolide on human pancreatic cancer cell line PANC-1 cells proliferation and apoptosis and its possible mechanism.Methods PANC-1 cells were treated in vetro with different concentrations of triptolide for different times.The inhibition of cells growth was evaluated by MTT assay.Flow cytometry was used to detect cellular apoptosis.RT-PCR and Western blot were used for detecting the expression of heat shock protein 70(HSP70).Caspase-3 activity was examined by Caspase-3 colorimetric assay.Results Triptolide significantly inhibited the proliferation of PANC-1 cells and induced apoptosis in a dose-dependent manner(P0.05).The HSP70 mRNA and protein expressions were inhibited dose-dependently by triptolide.Meantime,Caspase-3 activity in cytoplasm was elevated as the dose of triptolide increased(P0.05).ConclusionTriptolide in vetro can inhibit the proliferation of human pancreatic cancer PANC-1 cells and induce cell and apoptosis,which might be related to an inhibition of HSP70 expression and increase of Caspase-3 activity.
