Title: Plant regeneration of Muskmelon cv.Huanghemi
Abstract: This experiment was designed for plant regeneration from cotyledon and hypocotyls of muskmelon (Cucumis melo L.)cv.‘Huanghemi’.The results showed that the frequency of shoot regeneration of cotyledon explants was higher than that of hypocotyls explants,and the average shoot number regenerated from per cotyledon explants was more than that from per hypocotyls explants.The maximum shoot regeneration from cotyledon was obtained in MS containing 1.5 mg/L 6-BA and 0.2 mg/L NAA,the frequency was 60% and the average number of shoots per cotyledon was 9.4.The maximum shoot regeneration from hypocotyls was obtained in MS containing 1.75 mg/L 6-BA and 0.2 mg/L NAA,the frequency was 9% and the average number of shoots per hypocotyls was 8.5.After dark culture for 2 days,the roots was regenerated in 1/2 MS medium containing 0.2 mg/L IAA in the highest frequency,the frequency was 91.12% and the average number of roots per shoot was 12.Irrigation with 1/2 MS solution after socking in 200 mg/L NAA solution for 20 min was a good way for rooting out of the tuber,and the frequency was 52%.
