Title: Expression of SGK1 in the Renal Interstitium of Obstructive Nephropathy and Effect of Dracohodin Perchlorate on SGK1
Abstract: Objective To investigate the dynamic expression of serum and glucocorticoid induced kinase 1(SGK1) in rat renal interstitium of unilateral ureteral obstruction(UUO) and the effect of Dracohodin Perchlorate(DP) on the expression of SGK1.Methods The model of rat renal interstitial fibrosis was established by unilateral ureteral obstruction.Sixty Sprague-Dawley rats were randomly divided into four groups which were sham operation group,model group,DP treatment groups with low and high dose.Five rats of each group were sacrificed at 3,7 and 14 days after surgery,respectively.The levels of blood urea nitrogen(BUN)and serum creatinine(SCr) were determined.Renal interstitial fibrosis was confirmed by PAS staining and the expression of renal interstitium SGK1 protein was detected by immunohistochemistry assay.Results Compared to rats in sham operation group the levels of BUN and SCr were increased significantly with the development of interstitial fibrosis at 7 and 14 days in rats of model group(P0.05).Compared to rats of model group renal function and interstitial fibrosis were improved significantly at 7 and 14 days in rats of high dose DP treatment group(P0.05).The expression of SGK1 protein in renal interstitium of rats in model group was up-regulated and enhanced progressively in time(P0.05).However,the expression of SGK1 protein in renal interstitium of rats in high dose DP treatment group was reduced significantly in contrast to model group(P0.05).Conclusion The expression of renal SGK1 is increased with the progression of UUO,which suggests that SGK1 may play an important role in the development of obstructive nephropathy.Dracohodin Perchlorate probably could prevent renal interstitium fibrosis by inhibiting the expression of SGK1.
