Title: Purification of Serum IgG from Meriones Unguieulataus and the Production of Rabbit Polyclonal Antiserum
Abstract: Objective To purify serum IgG from Meriones unguieulataus with high purity and produce polyclonal rabbit antiserum against M.unguieulataus IgG.Methods The IgG from serum M.unguieulataus was purified by Hitrap protein-G affinity chromatography.The purity of the M.unguieulataus IgG was identified by SDS-PAGE and Western blotting,The M.unguieulataus IgG was subjected to immunize the rabbits to produce the antiserum.Results Using protein-G affinity chromatography method,7 mg IgG of the M.Unguieulataus was obtained from 7 mL M.unguieulataus's serum.And the purified IgG was showed high purity on SDS-PAGE and Western blotting analysis.The polyclonal rabbit antiserum against M.unguieulataus IgG was produced by immunizing the rabbit and the titer of the antiserum was up to 1∶16 on Double Immunodiffusion.Conclusion The method was established for purification IgG fraction from M.unguieulataus's serum and the polyclonal rabbit antiserum against M.unguieulataus IgG was obtained.Our results also suggested that M.unguieulataus IgG has high binding activity with Protein G.
