Title: Identification of Antifungal Strain A-29 and Its Active Components Analysis
Abstract: We isolated antifungal stain A-29 from the tomato roots in the soil of Shandong Academy of Sciences. We obtained the 16S rDNA sequence of 1426 bp by PCR amplification with the primers of 27f and 1492r,and the accession number EU430264 in GenBank. The BLAST search result showed that the highest 16S rDNA gene sequence homology between stain A-29 and Streptomyces viridocyaneus YIM30997 (AF513223) was approximately 98.9%.Phenotypic characteristics between A-29 and S. viridocyaneus were identical except xylose utilization and indole reaction,so strain A-29 was identified as Streptomyces viridocyaneus. The sterile fermentation extraction of strain A-29 had antifungal activity against such 8 species plant pathogenic fungi on a PDA dish as Pyricularia grisea,Verticillium dahliae,Colletotrichum lagenarium,Macrophoma kawatsukai,Bipolaris sorokinana,Gaeumannomyces graminis,Alternaria brassica,Fusarium graminearum. It had the best inhibitory effect against Alternaria brassica with 7.0 mm inhibitory radius. We employed Methylene dichloride or ethyl acetate to obtain the extracts from sterile fermentation extraction,which were analyzed with a Gas Chromatography-Mass Spectrometry (GC-MS). We deduced that the predominant components were,(S)-(+)-2′,3′-Dideoxyribonolactone,2-Vinyl-9-[3-deoxy-.beta.-d-ribofuranosyl]hypoxanthine ,2-(Phenyl-piperidin-1-yl-methyl)-cyclohexanol and10-Methoxy-nb-.alpha.-Methylcorynantheol after referring to other relevant literature.
