Title: The impact of STAT3 activation on bcl-2/bax expression and cell apoptosis in rats with acute lung injury
Abstract: Objective To investigate whether the activation of signal transducer and activator of transduction-3(STAT3)is protective against cell apoptosis and the mechanism underlying such effects in rats with acute lung injury.Methods Thirty healthy Wistar rats were randomized into 3 groups:normal saline(NS)group,lipopolysaccharide(LPS)group and Rapamycin(RPM,STAT3 specific inhibitor)pretreatment group.STAT3 mRNA expression was determined using in-situ hybridization;cell apoptosis and expression of bcl-2/bax were assessed using immunohistochemistry and TUNEL staining techniques.Results After Rapamycin inhibition of STAT3 mRNA expression,down-regulation of bcl-2 expression and up-regulation of bax expression was found in lung tissue.Meanwhile,an increase in cell apoptosis was also observed.Conclusion The activation of STAT3 pathway in ALI may cause up-regulation in bcl-2 expression and down-regulation in bax expression,which is protective against ALI-induced apoptosis.
