Abstract:Five lignan compounds,sehisantherin A(I_a),B(II_a), C(III_a),D(IV_a)and E(V_a)have been isolated from the fruits of Hua-Zhong-Wu-Wei-Zi(Schisandra sphenantheraRehd et Wils)(Magnoliaceae).In preliminar...Five lignan compounds,sehisantherin A(I_a),B(II_a), C(III_a),D(IV_a)and E(V_a)have been isolated from the fruits of Hua-Zhong-Wu-Wei-Zi(Schisandra sphenantheraRehd et Wils)(Magnoliaceae).In preliminary clinical trials,schisantherin A,B,Cand D showed signifieant effect in lowering the serum glutamie pyruvic transaminase(SGPT)level of the patients suffering from chronic virus hepatitis.The same effect has also been observed in experimental abnormal high SGPTlevel animals induced by carbon tetrachloride.The physical properties and the NMR data of sohisantherin A,B,C,D and E aregiven in Tab.1 and 2.The structure of schisantherin A reported previously is revised as follows,namelythe position of methylenedioxy group should be at 12,13(as I_a or I_b)instead of at 13,14,based on the following evidences,i.e.the studies of I_a and its related compoundson nuclear Overhauser effect(NOE)(Tab.3 and 4),solvent effect of methoxy protons(Tab.5) and chemical degradations (Chatt I).For the same reason,the publishedstructures of γ-schisandrin and wuweizisu C should be VI and VII respectively.Thestructures of schisantherin B,C,D and E are established as II_a,III_a,IV_a and V_arespectively,whose conformations(shown as II_b, III_b, IV_b and V_b respectively) werethe same as schisantherin A(I_b).When this work was finished,a communication on the chemistry of gomisia A,B,and C isolated from Schisandra chinensis Baill was published by Japanese chemists.It seems tha~+, gomisin B and C may be identical with schisantherin B and Arespectively,but schisantherin C,D and E have never been reported in the literatureso far.Read More
