Abstract:Objective To investigate the protective role of exogenous L-arginine on rats under immunological stress.Method Animal model of immunological stress was induced by Escherichia coli endotoxin(LPS) with ...Objective To investigate the protective role of exogenous L-arginine on rats under immunological stress.Method Animal model of immunological stress was induced by Escherichia coli endotoxin(LPS) with intraperitoneal injection.Thirty-two rats were randomly divided into 4 groups:normal control group,saline paired group,immunological stress group(LPS group) and L-Arginine+LPS group(L-Arg+LPS).Results Immunological stress was induced by intraperitoneal injection LPS 4 mg/kg·bw in rats.Serum arginine concentrations of rats in L-Arg+LPS group were extremely significantly higher than that of the other groups(P0.01).The activities of ALT in LPS and L-Arg+LPS group were extremely significantly higher than the control group(P0.01).Compared with the control group,the activities of T-NOS in LPS and L-Arg+LPS group were significantly increased(P0.05),and the concentrations of NO and the activities of iNOS extremely significantly increased(P0.01).The results of immunohistochemistry showed that eNOS expression of liver in L-Arg+LPS group was extremely positive,while the remaining groups had no eNOS expression.iNOS expression in liver was moderately positive in LPS group,and slightly positive in L-Arg+LPS group.There was no iNOS expression in control and paired group.Compared with control group,apoptosis of liver cells in LPS and L-Arg+LPS group was extremely significantly increased(P0.01),and apoptosis of liver cells in L-Arg+LPS group was significantly lower than that of LPS group(P0.05).There was no significant difference between control and saline paired group(P0.05).Conclusion Supplementation with arginine for rats under immunological stress could inhibit the inflammation and apoptosis of liver cells caused by NO from iNOS catalytic generation,and reduced the damage of immunological stress.Read More
