Title: Protective Effect of Chemokine Blocker's on Renal Ischemia/Reperfusion Injury In Rats
Abstract: Objective:To research protective effect and mechanism of Chemokine Blocker's-Lysophosphatidic Acid(LPA)on renal ischemia reperfusion injury. Methods:Male Wistar rats weighing 230 g±20 g were randomly divided into three groups(n=10): sham operation group, control group and LPA group. The murine model of renal I/R injury was made.Perfusion the kidney after 4 hours ischemia. We monitored the renal function, the concentration of MCP-1 by ELISA method and the renal pathologic changes through microscope. At the same time,we observed LPA's impact on these indexes. Results:The MCP-1 content of kidney was little in the sham operation group, while it increased obviously in the model group. After we had applied LPA to the model group, the level of serum Cr and BUN respectively lowered(P0.01),the content of MCP1 decreased(P0.01),grade of renal tubule fell in pathomorphology(P0.01).Conclusion:It is proved that cytokines-MCP-1 play a role in the renal ischemia-reperfusion injury.LPA protect kidney from disorder by inhibiting monocyte chemoattractant protein-1 and their anti-inflammatory effect.
