Title: Breeding of higher L-lactic acid mutants by protoplast mutagenesis
Abstract: Using Lactobacillus.casei ZZ-L,the optimal conditions of the protoplast preparation were studied in this experiment.The result showed that 22h cultured Lactobacillus.casei ZZ-L as material,0.7mol/L NaCl and 0.4mg/ml lysozyme were the optimum conditions of the protoplast preparation in 110mins.In the optimum conditions,preparation rate was 89.5% while the regeneration rate was 47.7%.The protoplast was used as the material in the UV-mutagenesis method for inducement.Then through plate screening and flask screening,8 high-yielding mutant strain was obtained by the target of L-lactic acid.The result showed that the best strain was ZZ-06 of which L-lactic acid of production was 78.6g/L,increased by 20.7% in comparison with Lactobacillus.casei ZZ-L.L-lactic acid of production kept stable after 12 generations of reproduction,which displayed perfect genetic stability.
