Title: Partition and Purification ofPigment from Rose by Aqueous Two-phase extraction
Abstract: Partition and purification of Rose pigment by PEG 400/Na2SO4 aqueous two-phase system was investigated in this paper.Determined aqueous two phase system was consisted with 25% PEG 400 and 18% Na2SO4.Influence of temperature,volume of pigment crude extract,pH on extraction efficiency of pigment was investigated through experiments.The results showed that the main factor iMPact the extraction efficiency of pigment was the volume of pigment crude extract.The second and third factors were pH in solution and temperature.The optimum extraction conditions include the followings:pH being 2.0,leaching solution volume 1mL and temperature 30℃.In this condition,the extraction rate of pigment reached to 96.86%.Aqueous two-phase is an effective method to separate and purify the pigment of plants.
