Title: SPE-HPLC determination of omeprazole concentration in serum
Abstract: Objective To estabilish a solid phase extraction and high performance liquid chromatography(SPE-HPLC)method for determination of omeprazole in human serum. Methods Serum samples were prepared with C18 solid phase extraction column. The chromatographic separation was performed on a Agilent TC-C18 column(4.6×150 mm,5 μm). The mobile phase consisted of 0.1 mol·L-1ammonium acetate(pH=7.0,adjusted by ammonia water)-acetonitrile(69:31,V/V),flow rate was 1.0 mL·min-1 . The detection wavelength was 302 nm. Results The linear range for omeprazole was 0.005~2 μg·L-1 (r=0.9997). The average recoveries of high,middle and low three concentration were 104.8 %、104.2 % and 102.4%,RSD were both less than 10 %.The intra-day and inter-day RSDs were less than 8.27%. Conclusions The method is accurate ,sensitive and simple,and suitable for the study on pharmacokinetic and bioavailability of omeprazole in human.
