Title: Screen and Analysis of Important Differentially Expressed Genes in the Genes Expression Profiles of Oral Leukoplakia
Abstract: Objective:To screen an important pathogenic oral leukoplakia differentially expressed genes and analyse their associated biological information.Methods:The extraction of the total white mucosa RNA,synthesis of single-labeled Cy3 fluorescent tag Crna,and then containing 41000 Genes/ESTs sequence of Agilent all of gene chips to Ratio≥2 and Ratio≤0.5 for the threshold to filter differentially expressed genes;of the differentially expressed genes in line GO(Gene Ontology) functional classification and pathway analysis;using RT-PCR on the part of the differentially expressed genes for validation.Results:41000 Genes/ESTs sequence were screened to the 892 differentially expressed genes,of which 623 were up-regulated genes and 269 were down-regulated genes.GO analysis of these differentially expressed genes mainly related metabolism,cell structure and so on;pathway of 7 were obtained significantly change the channel,the 667 differentially expressed genes are known,there are nine genes in the pathway occurred over seven enrichment.Down regulated gene Cyp2b13 RT-PCR gene Tyms line test consistent with the microarray results.Conclusion:The incidence of oral leukoplakia were mainly Cyp2b13,Tyms,Casp3,Cc15,CXCL12,Cc124,Dhfr,Apoe,Alb gene which lead to abnomal expression of Caspase-3activation pathway,Adhesion activity of chemokine receptor pathway,E2F transcription factor regulatory pathway of DNA replication,G1/S phase transition-related regulatory pathway,lipoprotein metabolism pathway,arachidonic acid metabolic pathway cytochrome P450 metabolic pathway changes,resulting the occurrence of leukoplakia.
